While watching Duke vids on Youtube I see people dropping down a command line interface while playing duke. How is this done and is there any instructions available for this?

You mean the console on EDuke32? Just press the ~ key on your keyboard.

In the version of eduke32 that comes with DukePlus (and probably a bunch of other versions), it is customizable.
In this version, when the player resets the keyboard layout to Classic, the console access is reset to C. This makes things like DNCLIP not work that great so I'll typically set it back to the tilde. It can be set to most anything, so it's worth checking and/or upgrading your version of eduke32 as needed.
Edit: r9063 packaged with DukePlus 2.5.1, found under Options/Control Setup/Keyboard Setup.
